T7-SAGE transcriptome of cleft versus end bud epithelial cells of embryonic mouse salivary glands

(Submitter supplied) Mus musculus embryonic submandibular salivary glands from embryonic day 12.5 embryos were laser microdissected to isolate epithelial cells from regions adjacent to forming clefts or from the peripheral end bud region. The isolated region-specific samples were then subjected to T7-SAGE transcriptome analysis.

Identifying the genes involved in CD4+ T cell mediated-acute allograft rejection

(Submitter supplied) Acute allograft rejection is a leading cause for the failure of organ allotransplantation. Identifying the genes involved in the rejection process provides clues to study the mechanisms, and to provide specific gene targets for monitoring, predicting and preventing acute allograft rejection. Using a mice model of skin acute allograft rejection and SAGE method, we analyzed gene expression in the CD4+ T cells of the mice, the cell type known to play critical roles in acute allograft rejection. more...

Profiling of spatio-temporally regulated transcripts during cerebral corticogenesis in the mouse

(Submitter supplied) The cerebral cortex plays an important role in cognitive function and specialized perception in mammals and its development requires highly specific spatio-temporal control of gene expression. The study identified stage- and region-specific markers throughout cerebral corticogenesis at various important stages of cerebral cortex development; embryonic day (E) 15.5, E17.5, postnatal day (P) 1.5 and 4-6 months old.

Mus musculus hematopoietic stem cell

(Submitter supplied) A transcriptome study in mouse hematopoietic stem cells was performed using a sensitive SAGE method, in an attempt to detect medium and low abundant transcripts expressed in these cells. Among a total of 31,380 unique transcript, 17,326 (55%) known genes were detected, 14,054 (45%) low-copy transcripts that have no matches to currently known genes. 3,899 (23%) were alternatively spliced transcripts of the known genes and 3,754 (22%) represent anti-sense transcripts from known genes.

Gene expression variations caused by dietary changes in methy-donor content

(Submitter supplied) In this study we adapted and tested the applicability of Methylation Specific Digital Karyotyping (MSDK), a non-array based method, for the prospective analysis of epigenetic changes after perinatal nutritional modifications in a mouse model of allergic airway disease. As part of the same study we also analyzed the gene expression profiles by SAGE.

Regulation of pituitary gene expression by adrenalectomy

(Submitter supplied) Overproduction of adrenal hormones causes obesity and hypertension, whereas adrenalectomy (ADX) leads to weight loss and hypotension. The pituitary gland is a major target organ for the adrenal hormones. To identify potential mediators for the effect of ADX on pituitary system regulating weight loss and hypotension, we investigated the effect of ADX on pituitary gene expression using serial analysis of gene expression (SAGE). more...

Regulation of hypothalamic gene expression by glucocorticoid: implications for energy homeostasis

(Submitter supplied) The current study has investigated the hypothalamic gene expressions regulated by glucocorticoid (GC), key hormones in energy homeostasis. Using serial analysis of gene expression (SAGE) method, we have studied the effects of adrenalectomy (ADX) and GC on the transcriptomes of mouse hypothalamus. Approximately, 180 000 SAGE tags, which correspond to 50 000 tag species, were isolated from each group of intact or adrenalectomized mice, as well as 1, 3 and 24 hours after GC injection. more...

SAGE in Cultured Murine COC During Early Maturation

(Submitter supplied) In cultured cumulus oocyte complexes (COC) FSH induces gene transcription required for germinal vesicle breakdown (GVBD). Experiments were performed to determine the critical period when gene transcription is required for GVBD and identify candidate mRNAs involved. Experiment I: murine COC were cultured 4h in the presence of FSH with 5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole (DRB) added at different intervals after the start of culture. more...

Signalling molecules in MEFs/FL

(Submitter supplied) In silico analysis identified signalling molecules related to Wnt (Wnt1, 2, 5a, 5b, 6, 11, 14, Wisp1, 2, sFRP1 and 2, Dkks 2 and 3) and BMP (BMP1, 4, 5) pathways. Many of these tags matched multiple transcripts in GenBank, while other potential molecules could not be detected because the transcripts either lacked an anchoring enzyme site or the site was immediately adjacent to the polyA+ sequence. Sexually dimorphic gene expression in the hypothalamus, pituitary gland and cortex by SAGE

(Submitter supplied) C57BL6 mice (12-15 weeks old) were obtained from Charles River Laboratories (St. Constant, Québec). They were housed in an air-conditioned room (19-25Åé) with controlled lighting from 07:15 to 19:15 h and were given free access to laboratory chow (Lab Rodent Diet No. 5002, Ren’s Feed and Suppliers, Ontario) and water. The hypothalamus, pituitary gland and parietal cortex of 51 male and 51 randomly estrous cycling female mice were pooled and used for expression profiling. more...
