Analysis of previously reported BRD4 target genes related to mitochondrial function in senescent fibroblasts.

(Submitter supplied) Because BRD4, a major target of ARV825, has been implicated in the regulation of mitochondrial gene expression in cardiomyocytes (DOI: 10.1161/CIRCULATIONAHA.120.047239), we examined whether ARV825-induced cell death in senescent cells is attributable to changes in the expression of mitochondria-related genes. Although senescent cells were eliminated within two days of ARV825 treatment, the mRNA expression levels of mitochondrial-related genes remained unchanged. more...

The hegemonic EWSR1::ETS oncogene overrules core regulatory circuitry mechanisms in Ewing sarcoma [ChIP-seq 2]

(Submitter supplied) Ewing sarcoma is an aggressive bone tumor of adolescence characterized by a hallmark EWSR1::FLI1 fusion oncogene. This chimeric protein drives tumorigenesis by reshaping transcriptional and epigenetic landscapes. However, how it is transcriptionally regulated and whether additional master transcription factors (MTFs) form a core regulatory circuit (CRC) in Ewing sarcoma remain unclear. Here, using an extensive panel of Ewing sarcoma cell lines and primary tumors, we mapped super-enhancers and identified strong enrichment of GGAA microsatellites, confirming their specificity to Ewing sarcoma as compared to other pediatric cancers and normal tissues. more...

Up-regulation of an epithelial miRNA is associated with immune evasion in progressive bronchial premalignant lesions [PCGA_miRNA_BX]

(Submitter supplied) Bronchial premalignant lesions (PMLs), precursors of lung squamous cell carcinoma, have distinct molecular subtypes. The proliferative subtype, enriched with bronchial dysplasia, had decreased expression of an antigen processing/presentation gene co-expression module in progressive/persistent versus regressive PMLs, suggesting a functional impact of these genes on immune evasion. Here, we performed miRNA sequencing, miRNA in situ hybridization (ISH), and spatial proteomics of bronchial biopsies from patients at high risk for lung cancer. more...

Aging-associated Increase of GATA4 levels in Articular Cartilage is Linked to Impaired Regenerative Capacity of Chondrocytes and Osteoarthritis

(Submitter supplied) Although the causal association between aging and osteoarthritis (OA) has been documented, our understanding of the underlying mechanism remains incomplete. To define the regulatory molecules governing chondrocyte aging, we performed transcriptomic analysis of young and old human chondrocytes from healthy donors. The data predicted that GATA binding protein 4 (GATA4) may play a key role in mediating the difference between young and old chondrocytes. more...

RNA sequencing of FZD6 knockdown in prostate cancer cells

(Submitter supplied) Wnt signaling is a complex pathway consisting of numerous ligands and frizzled (FZD) receptors. These signaling components are widely expressed in human prostate tissues and often undergo upregulation or mutation in advanced prostate cancers. Enhanced Wnt signaling promotes prostate cancer cell proliferation, metastasis, and resistance to therapy. However, targeting pan-Wnt signaling poses challenges due to tissue toxicity. more...

Niclosamide synergizes with sunitinib against renal cell carcinoma via regulation of DNA repair and cell cycle pathways

(Submitter supplied) Tyrosine kinase inhibitors (TKIs), such as sunitinib and sorafenib, are standard treatments for renal cell carcinoma (RCC). However, most patients treated with these drugs eventually develop drug resistance and relapse; therefore, new treatment options for RCC are urgently required. Recent studies have focused on combination therapies targeting distinct molecular pathways that may produce synergistic effects and help overcome drug resistance in RCC. more...

CUEDC1 promotes glycolytic metabolism reprogramming through the CUEDC1/CACNG4/PI3K axis to promote ER-positive breast cancer growth

(Submitter supplied) Background The reprogramming of energy metabolism, particularly glycolysis, upholds the malignancy of tumors. The relationship between CUE Domain Containing protein-1 (CUEDC1) and glycolysis, along with its influence on the development of ER-positive breast cancer (ER+ BRCA), is not well defined. This investigation explores the functional involvement of CUEDC1 in glycolysis regulation and uncovers a previously unidentified pathway contributing to the progression of ER+ BRCA. more...

T Cell Transcriptome Alterations in Healthy Volunteers Exposed to Three Weeks of Simulated Microgravity

(Submitter supplied) Impairment or reduction of lymphocyte-mediated immune response has been observed during space flight or in microgravity simulation conditions, with the underlying mechanisms remaining unclear. T lymphocytes (T cells) play a critical role in human immune response. The immunosuppression encountered by humans in microgravity may be linked to its impact on T cell global transcriptome networks, affecting immune cell-cell interactions. more...

Genetic alterations of SENP6 disrupt DNA repair and proteostasis conferring sensitivity to proteasome inhibition in multiple myeloma

(Submitter supplied) Targeting of the ubiquitin-proteasome system (UPS) remains a mainstay in the treatment of multiple myeloma (MM). Hyperactivation of the closely related SUMO pathway is implicated in disease progression and associated with poor prognosis. Here we investigated the functional imbalance of aberrant SUMOylation in MM by analyzing the large CoMMpass dataset for molecular alterations associated with SUMO pathway regulation. more...

Autosomal recessive RDH12-Leber congenital amaurosis retinal organoids display reduced photoreceptor length and phospholipid dysregulation

(Submitter supplied) NADPH-dependent retinol dehydrogenase 12 (RDH12) reduces all-trans-retinal in photoreceptor inner segments to regulate the phototransduction cascade and control the concentration of retinoids in the retina. Biallelic variants in RDH12 have been reported to cause autosomal recessive (AR) Leber congenital amaurosis (LCA), a severe cause of childhood blindness. Animal models have failed to recapitulate the clinical phenotype, hence, we generated human induced pluripotent stem cell (hiPSC) derived retinal organoids (RO) from the dermal fibroblasts of a female patient with homozygous missense variant c.619A>G p.(Asn207Asp) in RDH12 (RDH12-AR), and an unrelated healthy control (WT). more...
