Role of PLK1 in epithelial-mesenchymal transition in human melanoma

(Submitter supplied) Polo-like kinase 1 (PLK1), a serine/threonine kinase and important cell cycle regulator, is overexpressed in melanoma and its expression has been associated with poor disease prognosis. PLK1 has been shown to interact with NUMB, a NOTCH antagonist. However, the exact role of PLK1-NUMB-NOTCH axis in epithelial-mesenchymal transition (EMT) and melanoma progression is unclear. In this study, Affymetrix microarray analysis was performed to determine differentially expressed genes following doxycyclin induced shRNA-mediated knockdown of PLK1 in human melanoma cells that showed significant modulations in EMT and metastasis related genes.

Gene expression analysis of PBMC's from IBD subjects stimulated by combinations of LPS, MDP, anti-CD3/anti-CD28 antibodies, and anti-IL10R antibodies.

(Submitter supplied) We systematically investigated the transcriptomic response pattern of PBMC from 41 patients with IBD to characterize relevant stimuli that target different aspects of innate and adaptive immune cell responses. Lipopolysaccharide(LPS), muramyl-dipeptide (L18MDP), T cell receptor and co-stimulation (αCD3/αCD28 coated beads), and IL-10 signalling blockade were used to sitmulate subject PBMC's alone or in combination based on the concept that innate pathogen recognition receptor responses (in particular NOD2 and TLR4), T cell responses, and IL-10 signalling defects are implicated by multiple genetic IBD susceptibility loci and Mendelian forms of IBD.

Gene expression and alternative splicing profile of MFAP1- and THOC1-depleted human cells

Here we show that human THO interacts with MFAP1, a spliceosome-associated factor.
