Gene expression analysis of human keratinocytes (HaCaTs) after non-thermal plasma treatment

(Submitter supplied) Investigation of gene expression in human skin keratinocytes (HaCaT) following non-thermal plasma treatment for 20 s, 60 s, and 180 s compared to untreated and H2O2-treated controls. Microarrays were used to analyze and investigate the biological effects of non-thermal plasma on human keratinocyte cells. Using an argon plasma jet kinpen, regulated transcripts were analyzed and further described in Schmidt et al. more...

A genome-wide analysis of open chromatin in human tracheal epithelial cells reveals novel candidate regulatory elements for lung function (array)

(Submitter supplied) Distal cell-type-specific regulatory elements may be located at very large distances from the genes that they control and are often hidden within intergenic regions or in introns of other genes. The development of methods that enable mapping of regions of open chromatin genome wide has greatly advanced the identification and characterisation of these elements. Here we use DNase I hypersensitivity mapping followed by deep sequencing (DNase-seq) to generate a map of open chromatin in primary human tracheal epithelial (HTE) cells and use bioinformatic approaches to characterise the distribution of these sites within the genome and with respect to gene promoters, intronic and intergenic regions. more...

A genome-wide analysis of open chromatin in human epididymis epithelial cells reveals novel candidate regulatory elements for male infertility [Transcriptome data]

(Submitter supplied) The epithelium lining the epididymis has a pivotal role in ensuring a luminal environment that can support normal sperm maturation. Many of the individual genes that encode proteins involved in establishing the epididymal luminal fluid are well characterized. They include ion channels, ion exchangers, transporters and solute carriers. However, the molecular mechanisms that coordinate expression of these genes and modulate their activities in response to biological stimuli are less well understood. more...

Gene expression analysis of HaCaTs after non-thermal plasma treatment

(Submitter supplied) Investigation of gene expression in cultured human skin epithelial keratinocytes (HaCaT) following non-thermal plasma treatment for 60 s, compared to untreated control. Non-thermal atmospheric pressure plasma has recently gained attention in the field of biomedical and clinical applications. In the area of plasma medicine research one promising approach is to promote wound healing by stimulation of cells involved. more...

Histone variant H2A.Bbd is associated with active transcription and mRNA processing in human cells [mRNA]

(Submitter supplied) Variation in chromatin composition and organization often reflects differences in genome function. Histone variants, for example, replace canonical histones to contribute to regulation of numerous nuclear processes including transcription, DNA repair and chromosome segregation. Here we focus on H2A.Bbd, a rapidly evolving variant found in mammals but not in invertebrates. We report that in human cells, nucleosomes bearing H2A.Bbd form unconventional chromatin structures enriched within actively transcribed genes and characterized by shorter DNA protection and nucleosome spacing. more...

array-CGH profiling of human neuroblastoma samples obtained from infants included in the INES99.1, INES99.2 and INES99.3 trials

(Submitter supplied) In neuroblastoma (NB), the presence of segmental chromosome alterations (SCA) is associated with a higher risk of relapse, even when occurring together with numerical chromosome alterations (NCA). Furthermore, recent evidence shows that SCAs play a role in tumor progression. In order to analyze the role of SCAs in infants with NB, we have performed an extensive retrospective array-CGH analysis of tumours from infants enrolled in the European INES 99.1, 99.2 and 99.3 trials. more...

Expression analysis of human adipose tissue stem cells (ADSCs) transduced with homeobox C8 (Hoxc8) or control vector, followed by treatment with adipogenic-induction media for 14 days

(Submitter supplied) Adipogenesis occurs through a specific gene program in undifferentiated fat progenitors. We hypothesized that the properties of the fat progenitors are regulated by hox genes, the developmental genes essential in different tissue stem cells. Their biased expression in white and brown fat implies roles in distinguishing the two fat types. Among 39 Hox genes, Hoxc8 is highly enriched in undifferentiated adipose tissue stem cells (ADSCs) and down-regulated in differentiated adipocytes. more...

Expression Profiling of Androgen Receptor and Vitamin D Receptor Mediated Signaling in Prostate Cancer Cells

(Submitter supplied) Studies of gene expression profiles using the whole genomewide microarray analysis in LNCaP cells (AR+, p53wt) when treated with 5nM testosterone and 100nM 1,25(OH)2D3 alone or in combination. Comparisons between each treatment groups provide evidence for the crosstalk between VDR and AR mediated signaling events at the transcriptional levels, which may have significant clinical impact in patient care.
