Effect of Sin3a deletion on gene expression within FoxP3+ T-regulatory cells

(Submitter supplied) Foxp3 (forkhead box protein 3) functions as the master transcriptional regulator of Treg phenotype. Histone deacetylases 1 and 2 (HDAC1/2) contribute to the regulation of FoxP3 expression via interactions with a myriad of co-regulatory factors. While the nuclear scaffolding protein, Sin3a, has been well established as a co-factor of HDAC1/2, its role within FoxP3+ Tregs has not been. We analyzed the effects of conditional deletion of Sin3a in Foxp3+ Treg cells using three orthogonal approaches. more...

Single-nuclei characterization of oxycodone's effects on dorsal peduncular nucleus

(Submitter supplied) In addition to their intrinsic rewarding properties. opioids can also evoke aversive reactions that protect against misuse. Cellular mechanisms that govern the interplay between opioid reward and aversion are poorly understood. We used whole-brain activity mapping to show that neurons in the dorsal peduncular nucleus (DPn) are highly responsive to the opioid oxycodone. Connectomic profiling revealed that DPn neurons innervate the parabrachial nucleus (PBn). more...

KAS-seq profiling captures transcription dynamics during oocyte maturation

(Submitter supplied) In fully grown oocytes, the genome is considered to be globally transcriptionally silenced. However, this conclusion is primarily derived from the results obtained through immunofluorescence staining or inferred from the highly condensed state of chromosomes, lacking more direct evidence. Here, by using a kethoxal-assisted single-stranded DNA sequencing (KAS-seq) approach, we investigated the landscape of single-strand DNA (ssDNA) throughout the genome and provided a readout of the activity and dynamics of transcription during oocyte meiotic maturation. more...

RNA-SEQ for different CD8 T cell subsets of leukemic mice before and after chemotherapy

(Submitter supplied) Purpose: The origin of CD8 in bone marrow after AML chemotherapy is still unclear. To solve this problem, we collected CD8 cells after the onset of disease and chemotherapy for RNA sequencing. Methods:CD8 from bone marrow and tissue of mice after disease onset and chemotherapy were collected, sorted and sequenced. Results:After chemotherapy, CD8 undergoes migration and phenotypic transformation home to the bone marrow.

Neutrophil trapping and nexocytosis are mast cell processes for inflammatory signal relay

(Submitter supplied) Neutrophils are sentinel immune cells with essential roles for our anti-microbial defense. Most of our knowledge on neutrophil trafficking and tissue navigation derived from models of sterile inflammation and infection. Here, we analyzed allergen-challenged mouse tissues and discovered that degranulating mast cells (MCs) trap living neutrophils inside them. MCs release the attractant leukotriene B4 to re-route neutrophils toward them, thus exploiting a chemotactic system which neutrophils normally use for intercellular communication. more...

Gene expression of pdgfrb+ cells in the intact and injured spinal cord.

(Submitter supplied) Spinal cord injury (SCI) leads to fibrotic scar formation at the lesion site, which finally affects axon regeneration and motor functional recovery. Myofibroblasts have been regarded as the main cell types that filled in the fibrotic scar, however, the cell source of myofibroblasts in transection and crush SCI model remain to be elusive. Here we used lineage tracing or single cell transcription sequencing to investigate the cell origin of fibrotic scar. more...

Mast cells trap and cannibalize swarming neutrophils for metabolic independence

(Submitter supplied) Neutrophils are activated and attracted to sites of anaphylaxis by degranulating tissue resident mast cells. After they facilitate inflammation and an anti-microbial immune response. However, the dynamics of neutrophils and direct interactions with mast cells are poorly investigated yet. By approaching 2-photon intravital microscopy and live cell imaging, we show that degranulating mast cells induce neutrophil swarming by releasing leukotriene B4 (LTB4). more...

The Capicua C1 Domain is Required for Full Activity of the CIC::DUX4 Fusion Oncoprotein

(Submitter supplied) Rearrangements between genes can yield neomorphic fusions that drive oncogenesis. Fusion oncogenes are made up of fractional segments of the partner genes that comprise them, with each partner potentially contributing some of its own function to the nascent fusion oncoprotein. Clinically, fusion oncoproteins driving one diagnostic entity are typically clustered into a single molecular subset and are often treated a similar fashion. more...

Transcriptome sequencing of hippocampal tissue of APP/PS1 mice treated with rotating magnetic field for 12 months

(Submitter supplied) Amyloid beta (Aβ) monomers aggregate to form fibrils and amyloid plaques, which is one of the important mechanisms in the pathogenesis of Alzheimer's disease (AD). Since the Aβ1-42 aggregation has prominent role in plaque formation, which eventually lead to the patient's brain lesions and cognitive impairment, an increasing number of studies have been devoted to reduce Aβ aggregation process to slow down AD progression. more...

TMEM232 is required for the formation of sperm flagellum and male fertility in mice

(Submitter supplied) Asthenoteratozoospermia is a major cause of male infertility. Thus far, the identified related genes can explain only a small share of asthenoteratozoospermia cases, suggesting the involvement of other genes. The transmembrane protein TMEM232 is highly expressed in mouse testes. In the present study, to determine its function of TMEM232 in testes, we constructed a Tmem232-null mouse model using CRISPR–Cas9 technology. more...

The target gene investigation in osteoclats (OCs) treated with mmu-miR-5112 enriched in EVs from OCs with prostate cancer (PCa) cells [target_EV-miR-5112]

(Submitter supplied) Bone metastatic lesions of PCa contain areas of bone destruction and formation that are directed by OCs and osteoblasts (OBs), respectively. OC-derived EVs are reported to maintain normal bone homeostasis; however, there is no study investigating the role of EVs from OCs in the presence of PCa. We identified mmu-miR-5112 enriched in EVs from OCs co-cultured with several PCa cell lines. NGS analyses were performed to investigate predicted target genes of EV-mmu-miR-5112 in OCs.

The gene expression profiles of maturing osteoclasts (OCs) co-cultured with prostate cancer (PCa) cell lines.

(Submitter supplied) Bone metastatic lesions of PCa contain areas of bone destruction and formation that are directed by OCs and osteoblasts (OBs), respectively. Bone resorption induced by OCs is a necessary priming event for tumor progression in PCa bone metastasis.; however, the characteristics of OCs affected by PCa have not been fully elucidated. Here, we performed NGS analysis to investigate the characteristics of OCs in the presence of PCa.
