Identification of target genes of tumor suppressor microRNAs in human cancer

(Submitter supplied) To identify target genes of tumor suppressive microRNAs in human cancer, several cell lines (bladder cancer, prostate cancer, renal cell carcinoma, and head and neck squamous cell carcinoma) were subjected to Agilent whole genome microarrays. miR-517a, miR-218, miR-145, miR-1 and miR-874 function as tumor suppressors.

Identification of miR-145 or miR-133a target genes in bladder cancer

(Submitter supplied) We recently identified a subset of down-regulated miRNAs such as miR-145 and miR-133a in bladder cancer. Cell growth inhibitions occurred in miR-145 and miR-133a transfectants compared with the controls, suggesting that both miRNAs function as tumor suppressors. The aims of our expression studies were identification of these miRNAs target genes.

Cytoskeletal TAGLN2 is involved in gender-dependent adipose tissue expandability

(Submitter supplied) The preadipocytes’ shape changes dramatically during differentiation, in parallel with lipid accumulation and cytoskeletal reorganization. Failure to do so may impact the flexibility of adipose tissue (AT) to shift between lipid storage and mobilization, leading to impaired metabolism and insulin resistance. Transgelin 2 (TAGLN2) was identified as a cytoskeletal protein expressed in AT and associated with obesity and inflammation both in mice and humans, being normalized upon weight loss. more...

MiR-133b regulates the expression of the actin protein TAGLN2 during oocyte growth and maturation: a potential target for infertility therapy

(Submitter supplied) In this study, the expression of miRNAs in metaphase I (MI) oocytes treated with or without insulin-like growth factor 1 (IGF-1) was observed by microRNA microarray analysis. Results show that 145 miRNAs were up-regulated and 200 miRNAs were down-regulated in MI oocytes after IGF-1 treatment. MiR-133b, which was up-regulated more than 30-fold, was chosen for further research.

EGI-1 cell line treated with Saracatinib vs EGI-1 cell line untreated; EGI-1 xenograft treated with Saracatinib vs EGI-1 untreated

(Submitter supplied) Biliary tract carcinoma (BTC) has a poor prognosis due to limited treatment options. There is therefore urgent need to identify new targets and to design innovative therapeutic approaches. Among potential candidate molecules, we evaluated the non-receptor tyrosine kinase Src, observing promising antitumor effects of its small molecule inhibitor Saracatinib in BTC preclinical models. The presence of an active Src protein was investigated by immunohistochemistry in 19 surgical samples from BTC patients. more...
