(Submitter supplied) Peripheral blood was acquired from the Pécs Regional Blood Transfusion Center (Pécs, Hungary) and processed using Lymphocyte Separation Medium 1077 (Promocell, Heidelberg, Germany) in accordance with the manufacturer's instructions. Subsequently, the viable mononuclear cells were isolated, counted, and seeded into 6-well plates at a density of 1,000,000 cells per well. The culture medium was specifically formulated using RPMI-1640 medium (Biowest, Nuaillé, France), enriched with 1% L-glutamine (Lonza, Basel, Switzerland), 2% penicillin/streptomycin (Lonza, Basel, Switzerland), and 10% exosome-depleted FBS (Thermo Fisher Scientific, Waltham, USA) to minimize EV interference from bovine serum, further supplemented with 100 ng/mL Peprotech M-CSF 300-25 (Gibco, Waltham, USA). more...

Polyploidy of MDA-MB-231 cells drives increased extravasation with enhanced cell-matrix adhesion

(Submitter supplied) Metastasis, the leading cause of cancer-related deaths, involves a complex cascade of events, including extravasation. Despite extensive research into metastasis, the mechanisms underlying extravasation remain unclear. Molecular targeted therapies have advanced cancer treatment, yet their efficacy is limited, prompting exploration into novel therapeutic targets. Here, we showed the association of polyploidy in MDA-MB-231 breast cancer cells and their extravasation, using microfluidic systems to reproduce the in vivo microvascular environment. more...

Transcriptional survey of the light-induced anthocyanin pathway in non-GM purple tomatoes

(Submitter supplied) Tomato MicroTom plants of the genotypes Aft/Aft x atv/atv (Aft/atv) and Aft/Aft x atv/atv x hp2/hp2 (Aft/atv/hp2) were used. Fruits showing purple anthocyanin pigmentation in the skin were collected at at the mature green (MG) and mature red (MR) stages to compare the transcriptomes of exocarp and mesocarp between the two developmental stages and the two genotypes with the objective to highlight the mechanisms through which light induces anthocyanin synthesis in tomato skin and impede it in mesocarp and other internal parts of the fruits.

Human visceral adipose tissue RT-PCR in individuals with obesity

(Submitter supplied) Visceral adiposity is more related with insulin resistance than BMI itself. To study the molecular signature of the subsequent evolution steps of the metabolic sequalae of obesity we collected omental adipose tissue samples from individuals with obesity while undergoing bariatric surgery. Participants (all with obesity) were divided in 4 groups according to glycemic profile: Insulin sensitive/normoglycemic group (IS NG) - OxHOMA2<1 and fasting glycemia < 100 mg/dL (n=17); Insulin resistant/normoglycemic group (IR NG) - OxHOMA2 > 1 and fasting glycemia < 100 mg/dL and HbA1c < 5.7% (n=24); Pre diabetes group (PD) - OxHOMA2 > 1 and fasting glycemia > 100 mg/dL or HbA1c [5.7% - 6.4%] (n=33); Type 2 diabetes group (T2D) - OxHOMA2 > 1 and fasting glycemia > 125 mg/dL or HbA1c > 6.4% (n=18). more...

Real-time quantitative PCR analysis of Lentinula edodes mycelium face on different stress treatments.

(Submitter supplied) L. edodes strain w1 was cultured at 25°C in MYG medium (containing 1% malt extract, 0.1% peptone, 0.1% yeast extract, and 2% glucose). Fresh mycelium blocks were cultured in a 9cm petri dish containing MYG medium. After 8-day dark cul- ture, and they were divided into different treatment groups (group A,B,C,D,E). Transcriptional profiling of biomass degradation-related genes during Herbivorax saccincola A7 growth on different carbon sources

(Submitter supplied) Herbivorax saccincola A7 is an alkali-thermophilic lignocellulolytic bacterium that was known to possess a strong xylan degradation ability. They can utilize a wide range of carbon sources, however the response and regulation mechanisms to different carbon sources, in terms of genes expression, are still not identified. In this study, H. saccincola A7 was cultured with cellobiose, cellulose, xylan, cellulose-xylan mixture, and empty fruit bunch (EFB) as the sole carbon source. more...

GeneQuery RNA Profile of Trigeminal Ganglia from Tumor Burden and Tumor Naïve Mice

(Submitter supplied) Cancer patients often experience changes in mental health, prompting an exploration into whether nerves infiltrating tumors contribute to these alterations by impacting brain functions. Using a mouse model for head and neck cancer and neuronal tracing we show that tumor-infiltrating nerves connect to distinct brain areas. The activation of this neuronal circuitry altered behaviors (decreased nest-building, increased latency to eat a cookie, and reduced wheel running). more...

Real-time quantitative PCR profiling of human miRNAs in CAG RNA-expressing cells

(Submitter supplied) We examined whether PAPD5 upregulation induced by mutant CAG RNA led to any dysregulation of cellular miRNA profiling. We found that, out of the 751 miRNAs examined, the expression of 186 miRNAs was reduced in EGFPCAG78-expressing human neuroblastoma SK-N-MC cells and was restored back to normal levels upon the knockdown of PAPD5 expression.

Real time quantitative PCR analysis of mouse biliary epithelial cells

(Submitter supplied) Biliary complications are disabling conditions that arise in up to 25% of liver transplanted patients, resulting in additional surgical procedures, re-transplantation or, in the absence of a suitable regraft, death. Here, we investigate the role of the primary cilia, a highly-specialised sensory organelle, in biliary injury leading to biliary complications. Human biopsies were used to study the structure and function of primary cilia in liver transplant recipients that develop biliary complications (N=7), compared to successful transplants (N=12). more...

Stratification of expression profiles of immune-related genes in the lesion of cutaneous leishmaniasis patients with L. major and L. tropica infection

(Submitter supplied) There is still no reliable human vaccination against cutaneous leishmaniasis (CL), a serious public health issue in many parts of underdeveloped nations like Morocco and Iran. There are few studies comparing the expression of immune-related genes in the skin lesions of CL patients infected with L. major and L. tropica. In this study, we used dcRT-MLPA to analyze the expression profiles of 144 immune response-related genes in CL patients from Morocco and Iran who had been exposed to L. more...

Characterization of Residual MicroRNAs in AAV Vector Batches Produced in HEK293 Mammalian Cells and Sf9 Insect Cells

(Submitter supplied) With more than 130 clinical trials and eight approved gene therapy products, AAVs stand as one of the most popular vehicles to deliver therapeutic DNA in vivo. One critical quality attribute analyzed in AAV batches is the presence of residual DNA, as it could pose genotoxic risks or induce immune responses. Surprisingly, the presence of small cell‑derived RNAs, such as micro‑RNAs, has not been previously investigated. more...
