Treatment protocol siRNA experiments were conducted by adding 50nM siRNA in optimem using RNAimax transfection reagent per manufacturers instructions. Cells were harvested at 72 hours post transfection for processing. For drug treatment experiments. 1uM EPZ-6438 or 1uM DMSO were added 3 days prior to transfections with siRNA as previously described.
Growth protocol Cells were grown in DMEM + 10% FBS + P +S and maintained for less than 10 passages.

Extraction protocol RNA was extracted using trizol and Direct-zol kit (Zymo Research) per manufacturers instructions
Libraries were constructed using Kapa stranded mRNA sequencing kit
