Extraction protocol Healthy foreskin tissue from circumcision (donors with no history of atopic dermatitis [AD] or inflammatory skin diseases) was used to isolate human primary keratinocytes (KCs). The tissues were preserved at 4℃ and transported to the laboratory on ice within 6 hours. After washing, the subcutaneous tissue was removed and the remaining tissue was cut into small pieces. The tissue pieces were digested with neutral protease at 4℃ overnight, followed by trypsin digestion with shaking at 37℃ for 7 minutes. Digestion was terminated with serum, and the cell suspension was filtered and centrifuged. Complete medium was prepared, and the cells were resuspended, seeded, and cultured at 37℃. Subsequent RNA extraction and sequencing were performed on KCs with SOCS6 knockdown or overexpression.

Data processing Raw reads were quality-controlled with fastp (v0.20.0), then aligned to the Homo sapiens reference genome (GRCh38) using HISAT2 (v2.2.1). Gene-level expression was quantified via featureCounts (v2.0.1) based on annotated genomic intervals, and transcript-level expression was estimated using StringTie (v2.1.6).
