RORγt -expressing FOXP3+ skin-resident Treg cells derived from the periphery in Allergen-Specific Immunotherapy

(Submitter supplied) The study explored the impact of Specific Immunotherapy on patients with Atopic Dermatitis sensitized to house dust mites by analyzing the transcriptomes of T regulatory cells before and after treatment. Result found that effective SIT is associated with distinct gene expression changes in Treg cells, particularly increased expression of Treg markers and RORC, a gene linked to immune regulation. These results indicate that successful SIT may enhance the function of Treg cells expressed RORC and help restore immune balance in these patients.

Expression data from T-cell Acute Lymphoblastic Leukemia (T-ALL) patient-derived xenograft treated with anti-CD3 mAbs.

(Submitter supplied) T-cell acute lymphoblastic leukemia is an aggressive hematological malignancy treated with chemotherapy and has a poor prognosis. In previous studies, we demonstrated that T cell receptor (TCR)-induced negative selection is highly effective in combating T-ALL in mouse models. Moreover, targeting the TCR of diagnostic T-ALL-derived PDX with anti-hCD3 (aCD3) mAb OKT3 or the clinically relevant aCD3 mAb Teplizumab resulted in leukemic cell death, regression of leukemia, and improved host survival. more...

Phenotypic Plasticity upon GSC Differentiation

(Submitter supplied) At the cellular level, the malignant characteristics of GBM are largely attributed to GBM stem cells (GSCs). These cells, endowed with stem-like properties, can self-renew, generate diverse cancerous cell populations, and initiate tumors in vivo. The GSC lines, NCH644 and NCH421k, when cultured as neurospheres (NS) — a condition that preserves their stem-like state, as evidenced by the expression of stem cell markers like SOX2, CD133, and Nestin — were found to align with the PN subclass and CL-B/C respectively.

Expression data from T47D/S2 (control) and T47D/TR-2 (tamoxifen-resistant) cell lines

(Submitter supplied) Basal expression profiling was performed to understand molecualr pathways specifically altered in tamoxifen-resistant cells. Tamoxifen is the most accepted targeted agent for treating patients with estrogen receptor (ER)-positive breast cancer (BC), but its efficacy has been limited due to drug resistance, manifested as disease recurrence and eventually death. Metabolic reprogramming of lipids has recently been recognized as a new mode of therapeutic resistance in various cancers. more...

Expression data from Hap1 parental control and Baz2B-KO cell lines

(Submitter supplied) Baz2B protein was shown to form a chromatin remodelig complex mit Snf2H or Snf2L catalytic subunits. It is known that chromatin remodeling proteins regulate diffferent DNA-templated processes, therefore we wanted to know whether Baz2B plays a role in transcription regulation. We used microarrays to detail the global expression of transcripts in Hap1 parental cell line and compare them to Baz2B-KO. We found up- and down-regulated trasncripts that could be associated with the decreased cellular proliferation in Baz2B-KO cell line

Transcriptomic analysis of SLAN+ vs SLAN- non-classical monocytes

(Submitter supplied) Monocytes can be classified into four different subsets: classic, intermediate and SLAN- and SLAN+ non-classical monocytes. SLAN+ monocytes are significantly reduced in MM patients compare to the frequency found in healthy adults (HA). Thus, we sought to analyze the transcriptome of SLAN- and SLAN+ cells to unveil the differences among both populations We used microarrays to detail the global programme of gene expression underlying the differences among both cell subsets We developed a 12-color combination of mAbs to evaluate the contribution of each individual marker for identifying and subsetting BM TAMs in patients with MM (N = 5): CD36-UV405, CD16-UV525, CD163-BV450, CD45-BV525, CD206-BV610, CD86-BV660, CD62L-BV763, cyCD68-FITC, SLAN-PE, HLADR-PerCPCy5.5, CD300e-APC, CD14-APCH7. more...

Targeting SUMOylation dependency in human cancer stem cells through a unique SAE2 motif revealed by chemical genomics

(Submitter supplied) Natural products (NP) encompass a rich source of bioactive chemical entities. Hence, NPs-derived compounds represent the majority of drugs currently approved to treat cancer. Here, we used human cancer stem cells (CSCs) in a chemical genomics campaign with NP chemical space to interrogate extracts from diverse strains of actinomycete for anti-cancer properties. We identified a compound (SCCRI025044), capable of selectively inhibiting human CSC function vs. more...
