Maternal Circulating Exosomal miR-185-5p as Non-invasive Biomarker for the Prediction of Recurrent Pregnancy Loss.

(Submitter supplied) Pregnancy losses usually happening suddenly and are irreversible, this study aims to find a predictive biomarker of the upcoming pregnancy loss. We collected serum exosomes from pregnant women prospectively from 6 to 8 weeks gestation and divided the patients into ongoing pregnancy (OP) group, sporadic pregnancy loss (SPL) group, and recurrent pregnancy loss (RPL) group according to the pregnancy progress and pregnancy history. more...

Plasma exosomal microRNAs associated with metabolism as early predictor of gestational diabetes mellitus

(Submitter supplied) The microRNA (miRNA) expression profile of plasma exosome in pregnant women complicated with gestational diabetes mellitus (GDM) has not been fully clarified. In this study, differentially expressed miRNAs in plasma exosomes were identified by high-throughput small RNA sequencing in 12 GDM and 12 normal glucose tolerance (NGT) pregnant women and validated in 102 GDM and 101 NGT pregnant women. A total of 22 exosomal miRNAs were found and five of them were verified by qRT-PCR. more...

Identification of exosomal microRNA panel as diagnostic and prognostic biomarker for small cell lung cancer [mRNAseq_BEAS2B]

(Submitter supplied) Small cell lung cancer (SCLC) has an exceptionally poor prognosis; as most of the cases are initially diagnosed as extensive disease with hematogenous metastasis. Therefore, the early diagnosis of SCLC is very important and may improve its prognosis. To investigate the feasibility of early diagnosis of SCLC, we examined exosomal microRNAs (miRNAs) present in serum obtained from patients with SCLC. Exosomes were isolated in serum from patients with SCLC and healthy individuals and were characterized using particle size and protein markers. more...

Circulating serum miRNAs as potential diagnostic biomarkers for oesophageal adenocarcinoma

(Submitter supplied) Exosomes were purified from 250 ul serum using ExoQuickTm. The presence of particles consistent in size with exosomes (60-150nm) was confirmed using a Nanosight LM10. miRNA was extracted from exosomes using an miRNeasy Serum/Plasma kit (Qiagen, #217184). miRNA was reversed transcribed using a TaqMan® microRNA Reverse Transcription Kit (Life technologies, #4366596). miRNA profiling was performed with a high throughput TaqMan® OpenArray® Human microRNA panel (Life technologies, #4461104). more...

Human exosomal placenta-specific miRNAs modulate expression of target genes in maternal immune cells during pregnancy

(Submitter supplied) We investigated placenta-specific miRNA miR-517a (miR-517a-3p) functions in Jurkat cells. For this purpose, we identified candidate target mRNAs of the placenta-specific miRNAs using DNA microarray analysis. Transfection of Jurkat cells with miR-517a significantly downregulated 123 genes. Among the 123 genes identified, we searched for potential direct targets of miR-517a using the online software MicroCosm Targets. more...

Expression profiling in exosomal miRNAs of diabetes, diabetes with coronary microcirculation disorder, and diabetes with coronary artery disease

(Submitter supplied) Due to the insidious onset and the difficulty to diagnose and intervene, it is particularly important to find the diagnostic markers of diabetic coronary microcirculation disorder (CMD) and therapeutic targets. Circulating exosomes, as endogenous exosomes derived from multiple tissue cells, mediate communication between cells and tissues, may serve as a key point for future diagnosis, prognosis and personalized treatment of CMD in diabetes. more...
