Title Microarray data from hUCBMSC to investigate the role of resveratrol in the restoration of monocrotophos induced cellular damages.
Organism Homo sapiens
Experiment type Expression profiling by array
Summary Analysis of human umbilical cord blood mesenchymal stem cell (hUCBMSC) derived neuronal cells exposed with (10 μM) concentration of resveratrol (RV),(50 ng/ml) concentrations of nerve growth factor(NGF) and (10 μM+50 ng/mL) concentrations of RV+NGF , respectively for 30 minutes . hUCBMSC were co-exposed with monocrotophos (MCP) and RV, MCP and NGF, MCP and RV+NGF before cell differentiation and after cell differentiation. Results provide insight into the role of RV in the restoration of MCP-induced cellular damages, thus proving to have a therapeutic potential against organophosphate pesticide-induced neurodegeneration.
We used microarrays to detail the global programme of gene expression underlying the neurotoxicity of MCP and neuroprotection of RV on of human cord blood derived mesenchymal stem cells (hCBMSCs). Differential gene expression studies identified the role of RV and NGF in survival, proliferation and neuronal differentiation of hCBMSCs. Microarray analysis also identified distinct classes of up-regulated genes during this process.

Overall design Total 10 samples were prepared from human umbilical cord blood mesenchymal stem cells (hUCBMSC) for RNA extraction and hybridization on Affymetrix microarrays. hUCBMSC without any exposure is taken as control samples (control). hUCBMSC were exposed with resveratrol (RV), nerve growth factor (NGF), and resveratrol + nerve growth factor (RV+NGF). To study the neurotoxicity of MCP and neuroprotection of RV before cell differentiation(pre exposed samples), hUCBMSC were co-exposed with monocrotophos (MCP) and RV (MCP pre exposed RV), MCP and NGF (MCP pre exposed NGF), MCP and RV+NGF (MCP pre exposed RV+NGF) .We also studied the effect of RV and NGF on hUCBMSC after cell differentiation (post exposed samples), hUCBMSC were co-exposed with monocrotophos (MCP) and RV (MCP post exposed RV), MCP and NGF (MCP post exposed NGF), MCP and RV+NGF (MCP post exposed RV+NGF).
