Title |
Genome-wide maps of chromatin state in WT c-Myc and K427R mutant lymphoma cells |
Organism |
Homo sapiens |
Experiment type |
Genome binding/occupancy profiling by high throughput sequencing
|
Summary |
We report the application of single-molecule-based sequencing technology for high-throughput profiling of c-Myc with core promoters of its target genes in mammalian cells. We performed ChIP sequencing with antibody to total c-Myc with core promoters of its target genes in WT c-Myc and K427R mutant group. Inspection of multiple individual traces and global analyses showed that the K427R mutant globally reduced association of c-Myc with core promoters of its target genes.

Overall design |
we precipitated chromatin with tag antibody to WT c-Myc and K427R mutant, and sequenced the precipitated material. K427 mutation reduced association of c-Myc with core promoters of its target genes
