Summary Here, we show unexpected and crucial metabolic nodes through TCA flux and glutaminolysis that modulate germ layer specification. Previous reports have shown that sustained glycolytic flux though MYC regulation is required for ectoderm specification. Our data support the finding that the germ layers have distinct metabolic profiles, but through investigation into the role of OXPHOS in germ layer specification.
 
Overall design Bulk RNA-Sequencing of H9-derived hPSCs differentiated to early trilineage (mesoderm, endoderm, and ectoderm) using a nutrient-balanced media formulation. Samples represent n = 5-7 of each lineage (hPSC, ME, EN, and EC).
