Summary |
We found that PC-3 cells consisted of rBC2LCN-positive and -negative subpopulations. To examine a global gene expression comparison between the rBC2LCN-positive and -negative subpopulations, DNA microarray was performed.

Overall design |
Human prostate cancer cell line, PC-3 cells, was maintained in F-12K medium (ATCC, 30-2004) supplemented with 10 % FBS (GIBCO, 12483-020) and 1 % Penstrep (SIGMA, I0516-5ML) at 37 °C and 5 % CO2. PC-3 cells were separated into rBC2LCN-positive and -negative subpopulations by live cell sorting using HiLyte Fluor 647-conjugated rBC2LCN. The total mRNAs were extracted from rBC2LCN-positive and -negative subpopulations using ISOGEN (NIPPON GENE). DNA microarray was performed by a SurePrint G3 Human GE Microarray Kit, 8 x 60 K Ver. 1 (Agilent, G4851A).
