Summary |
Under microscope, MH-S cells show a heterogenous population. By clonal dilution, we generated single cell colonies and found that a set of clones that secreted higher the IL-4Ra regulating protein (“High IL-4Ra activity clones”) and set of colonies secreted lower amount (“Low IL-4Ra activity clones”). We performed microarrays to detail the global gene expression analysis comparing high IL-4Ra activity and low IL-4Ra activity MH-S clones stimulated with or without LPS. At this time point, roughly 50 genes annotated as “extracellular” were significantly upregulated more than two-fold in LPS stimulated clones compared to unstimulated ones. Of these, only seven genes showed higher expression in the high IL-4Ra activity clones compared to low IL-4Ra activity clones.

Overall design |
Three high IL-4Ra activity and three low IL-4Ra activity MH-S clones stimulated with or without LPS for 16 hrs were selected for RNA extraction and hybridization on Affymetrix microarrays. Total 12 samples were analysed.
