Title |
Chromatin remodeling in bovine embryos indicates species-specific regulation of genome activation |
Organism |
Bos taurus |
Experiment type |
Genome binding/occupancy profiling by high throughput sequencing
|
Summary |
Characterization of genome-wide chromatin accessibility in bovine oocytes and preimplantation embryos using ATAC-seq
|
|
|
Overall design |
ATAC-seq libraries were prepared from bovine GV oocytes and in vitro produced preimplantation embryos (2-cell, 4-cell, 8-cell, and morula-stage embryos). For each developmental stage, ATAC-seq libraries were prepared for three biological replicates (cells derived from three separate oocyte collections). A subset of embryos from each collection were concurrently cultured in the presence of a transcriptional inhibitor (α-amanitin), to interrogate the causal relationship between embryonic transcription and chromatin remodeling.
