Title |
Transcriptional changes induced by Lamin A/C downregulation in the nuclei of E0771 breast cancer cells |
Organism |
Mus musculus |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
Major transcriptional changes (290 differentially expressed genes) take place upon downregulation of Lamin A/C in E0771 breast cancer cells
|
|
|
Overall design |
Lamin A and lamin C expression in E0771 breast cancer cells were downregulated by stable introduction of a lamin A/C shRNA construct: MISSIONĀ® TRC2 pLKO.5-puro Empty Vector (shControl), Sigma-Aldrich Cat# SHC201 or MISSIONĀ® shRNA plasmid DNA Lamin A/C, Sigma-Aldrich TRC# TRCN0000317672 (shLmna), resulting in an expected increase in nuclear deformability and squeezing capacities in this breast cancer cell model. RNA was isolated from both shControl and shLmna E0771 cells for transcriptome analysis via MARS-seq sequencing and bioinformatics analysis.
