Title Single guide RNA CRISPR /Cas(sg) editing 3 nucleotides and prime editing (pe) single nucleotide of CArG element in Tspan2 promoter effect on transcriptome in aortas
Organism Mus musculus
Experiment type Expression profiling by high throughput sequencing
Summary Aortas from sgCArG WT versus sg CArG homozygous mice at 12 wks and peCArGWT and peCArG homozygenous mutant mice at 8 weeks were isolated for total RNA extraction. RNA samples were then subjected to deeping sequencing to profile gene expression in aortas.

Overall design We will compare the transcriptome of aortas from sg CArG WT versus sg CArG mutant as well as peCArG WT versus peCArG mutant to determine the genes consistently regulated by CArG mutation of TSPAN2 gene in mice.
