Summary Myocardial Ischemia-reperfusion injury is a serious clinical problem that lacks effective therapy.However, the precise mechanism leading to myocardial I/R injury remains unclear.Our study defined the dynamic changes of the differentially expressed genes related myocardial I/R injury, and provide basis for comprehensive understanding of the disease at molecular level.
We investigated schemia-reperfusion –mediated gene expression alteration associated with the development of cardiac injury by microarray in a mouse model.
 
Overall design All mice were euthanized by an overdose of pentobarbital on hours 6, 24 and 72 of ischemia/reperfusion (I/R) treated. Total RNA was isolated with TRIzol (Invitrogen) from heart organs (n=4 per group) at each time point according to manufacturer’s instructions. Gene expression profiling was performed using Affymetrix GeneChip mouse Genome 430 2.0 array according to the manufacturer’s instructions (Affymetrix, Inc., Santa Clara, CA). On the GeneChip Mouse Genome 430 2.0 Array, over 45,000 probe sets analyze the expression level of over 39,000 transcripts and variants from over 34,000 well characterized mouse genes.
