Overall design |
P18T organoids were injected subcutaneously into Swiss-nude mice. When tumors were grown above 100mm3, animals were sacrified and xenografts were dissagregated and stained for Epcam+ and Fab072 (Lgr5). Within the alive Epcam+ compartment, 1000 cells of the 10% most positive Fab072 cells vs. Fab072- were FACS sorted in picoprofile lysis buffer. Two replicas were collected from two different xenografts. RNA was extracted and amplified with the picoprofile technique and retrotranscrived to cDNA. samples were hybridized to Affimetrix Microarray Chips.
