Title Transcriptomic characterization of FLC-mutant engineered human hepatocyte organoids

Summary We engineered human hepatocyte organoids to study the effect of different fibrolamellar carcinoma mutations. To study the effects and differences between the different mutations, we performed bulk RNA-sequencing to understand the transcriptomic changes.
 
Overall design Multiple clonal lines per genotype (DNAJB1-PRKACAfus, BAP1KO, PRKAR2AKO and double BAP1KO;PRKAR2AKO) were generated and replicates of parental wild type organoids are included, all were cultured in hepatocyte medium. For wild type organoids and double mutant BAP1KO;PRKAR2AKO organoids samples were also analyzed when cultured in ductal medium.
