Summary |
Cardiac resident MerTK+ macrophages exert multiple protective roles post-ischemic injury. To determine the potential reason for the protective role of MerTK+ cardiac macrophages, microarray was performed to identify gene expression profiles on isolated Trem2+, MHCII+, Lyve1+, and MerTK- macrophages from the heart tissue of WT mice post-IR. We investigated the potential reason for the protective role of MerTK+ cardiac macrophages in myocardial Ischemia reperfusion injuryby microarray.

Overall design |
Trem2+, MHCII+, Lyve1+, and MerTK- macrophages were isolated from the heart tissue of WT mice post-IR. Total RNA was isolated with TRIzol (Invitrogen) from macrophages according to manufacturer’s instructions. Gene expression profiling was performed using Affymetrix GeneChip mouse Genome 430 2.0 array according to the manufacturer’s instructions (Affymetrix, Inc., Santa Clara, CA). On the GeneChip Mouse Genome 430 2.0 Array, over 45,000 probe sets analyze the expression level of over 39,000 transcripts and variants from over 34,000 well characterized mouse genes.
