Title Epididymal white adipose tissue expression data from WT and Abhd15-ko mice on normal chow diet at refed state

Summary Abhd15 is mainly expressed in white adipose tissues and highly upregulated upon adipogenesis. Abhd15 expression is correlated with insulin resistance in obese humans, however its physiological function remains unknown. We used the microarray technology to gain insight into ABHD15’s physiological function by identifying dysregulated genes in eWAT from Abhd15-ko mice in comparison to WT mice.
We used microarrays to detail the global programme of gene expression in epididymal white adipose tissue and identified distinct dysregulated genes in response to refeeding
 
Overall design Male WT and Abhd15-ko mice on a clean C57BL/6J background were used. At 14 weeks of age, WT and Abhd15-ko mice were fasted overnight and refed with chow diet for 1 hour prior to harvesting. eWAT from WT and Abhd15-ko mice was collected and immediately snap frozen in liquid nitrogen. RNA was isolated for gene expression microarray analyses at the exon level (Affymetrix, Santa Clara, CA, USA). To isolate RNA, the frozen tissue samples were homogenized in TRIzol® reagent (Invitrogen/Life Technologies, Carlsbad, CA, USA) and processed based on the manufacturer’s instruction. Total RNA (200ng) from individual samples was then used for Affymetrix analysis. Terminal-labeled cDNA, hybridization to genome-wide Mouse Gene 2.0 ST Gene Chips and scanning of the arrays were carried out according to the manufacture’s indications (Affymetrix). Output primary row data was analyzed with Expression Console software (Affymetrix).
