Frozen leukemia bone marrow cells were thawed using standard conditions and 1 million cells were used for fixing and subsequent ChIP seq steps.

DNA was sheared from the fixed MLL-AF6 leukemia cells using sonication. Histone-DNA complexes enriched for H3K79me2 were isolated by immunoprecipitation with the Abcam anti-H3K79me2 specific antibody (Ab 3594)
ChIP DNA libraries were made following the Illumina ChIP-seq library preparation kit.
