Frozen leukemia bone marrow cells were thawed using standard conditions and 1 million cells were used for fixing and subsequent ChIP seq steps.

All cell lines were grown in RPMI 10%FBS supplemented with 2mM L-glutamine and penicillin/streptomycin and used directly for fixing/crosslinking for ChIP-seq

DNA was sheared from the fixed leukemia cell lines using sonication. Histone-DNA complexes enriched for H3K79me2 were isolated by immunoprecipitation with the Abcam anti-H3K79me2 specific antibody (Ab 3594)
ChIP DNA libraries were made following the Illumina ChIP-seq library preparation kit.
