Growth protocol H1 embryonic stem cells were cultured in mTESR1 medium. Cells were passaged every 5-6 days.
Extracted molecule genomic DNA
Extraction protocol H3K18ac ChIP: 20,000,000 Cells were resuspended in 450 ul of lysis buffer and incubated for 10 min on ice and immediately sonicated using Misonix cup-horn sonicator. 100 ul of the lysate (corresponding to 5,000,000 cells) were used for each immunoprecipitation with H3K18ac antibody; 10 ul of the lysate was used as input. After overnight reversal of crosslinking at 65°C, samples were treated with RNase A for 30 min at 37°C, proteinase K for 2 h at 56°C. DNA was subsequently purified using phenol/chloroform extraction and ethanol precipitation. DNA concentration was measured using Qubit (Life Technologies). Sequencing libraries were prepared with Ovation Ultralow DR Systems (NuGEN) according to the manufacturer's instructions.

Data processing Reads were aligned to the human hg19 genome assembly using bowtie-0.12.7 with parameters -r --offrate 6 -m 1 --best --strata -t -v 2
peaks were called based on the p value for Poisson distribution of enriched immunoprecipitated DNA relative to input. Significant BrdU peaks were defined as those windows with a p value<10e-4 and with two neighboring windows at the same significance. Significant H3K18ac peaks were defined as those windows with a p value<10e-3 and with two neighboring windows at the same significance.
