Treatment protocol On day 2, culture medium was replaced and cells were exposed to Atorvastatin (ATO) (10 µM) for 24 h.
Growth protocol Human prostate cancer cells (PC3 cells and LNCaP cells) were purchased from the American Type Culture Collection (Manassas, VA). All prostate cancer cell lines were cultured in RPMI medium containing 10% FBS. PC3 and LNCaP cells were seeded in culture dishes so that cells reached ~30% confluence on Day 2.
Extracted molecule total RNA
Extraction protocol Total RNA to be used in both mRNA and miRNA assays was isolated and purified using the Ribopure RNA isolation kit (Invitrogen, Grand Island, NY).
Label Cy3
Label protocol Total RNA samples were dephosphorylated, denatured, and end-labeled according to Agilent's protocols.
 
Hybridization protocol Labeled target was applied to Agilent miRNA 8x15K arrays (Design ID#029297) and hybridized overnight.
Scan protocol Slides were washed and scanned with an Agilent G2565 Microarray Scanner (Agilent Technologies, Santa Clara, CA) at 5 µm resolution.
Data processing Agilent Feature Extraction software was used to process the scanned images from arrays (gridding and feature intensity extraction) and the data generated for each probe on the array was analyzed with GeneSpring GX v7.3.1 software (Agilent Technologies, Santa Clara, CA).
