Treatment protocol After 7 days in vitro, mouse cortical neurons were treated with drug or vehicle. After 24 or 72 hrs drug treatement, total RNA was prepared.
Growth protocol Mouse cortical neurons were cultured from E13-15 mouse embryos for 7 days in vitro.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from mouse cortical neurons using Trizol reagent.
Label biotin
Label protocol RNA was labeled using the Affymetrix/Ambion WT Expression kit, combined with Affymetrix GeneChip WT terminal labeling kit.
 
Hybridization protocol Arrays for experiments (1) and (3) were washed and stained using the Affymetrix Gene Fluidics station 450. Hybridization was performed at 48˚C for 17 hours. Arrays for other experiments were run using the Affymetrix GeneTitan system. Steps subsequent to hybridization were performed with the Beckman Coulter FXP Target Express Robot. Hybridization was performed at 45˚C for 16 hours with the Affymetrix GeneTitan instrument, controlled using AGCC software.
Scan protocol GeneTitan arrays were scanned using the Affymetrix GeneTitan instrument, controlled by AGCC software. Individual arrays were scanned using the Affymetrix GeneChip Scanner 3000 7G Plus with autoloader.

Gene Expression data for dose response to Topotecan treatment of mouse cultured cortical neurons
Data processing Arrays were normalized and expression values determined using Affymetrix Expression Console. Linear RMA normalization was used for all experiments.
