Treatment protocol A549 cells were transfected with either control siRNA (D-001810-10-05, Dharmacon) or SETX siRNA (23064, Dharmacon) pools using the Lipofectamine™ RNAiMAX Transfection Reagent (Invitrogen). Transfections were performed according to the manufacturer’s instructions. 48 hours post transfection, the cells were infected with the Influenza A/Puerto Rico/8/34(ΔNS1) strain at an multiplicity of infection of 3. Wells of uninfected cells were used as controls for infection. Cells were collected for further processing 4 hours post infection. RNA and protein levels of SETX were quantified to determine knockdown efficiency.
