Treatment protocol We established a BAHD1 gene-specific knockdown cell line using the SilenciX® siRNA delivery system (Tebu-bio). This technology is based on EBV-replication, and ensures chromosome-like replication of an episome stably expressing either a shRNA against BAHD1 (silXBAHD1) or a control shRNA (silX-CT).
Growth protocol Cells were grown at 37°C, in a humidified 10% CO2 incubator, in Dulbecco's modified Eagle's medium with GlutaMAX (Gibco) supplied with 10%FBS (Gibco) and 70 ug/ml Hygromycin B (10687-010-Invitrogen).
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the RNeasy Mini Kit (Qiagen), from three biological replicates, according to the manufacturer's instructions. RNA was treated with DNase I to remove residual DNA and then cleaned with DNase Inactivation Reagent (Ambion® Inc.) RNA was quality checked an then store at -80°C.
Label biotin
Label protocol RNA was amplified with GeneChip® WT cDNA Synthesis and Amplification Kit (Affymetrix) and biotin labeld with GeneChip® WT Terminal Labeling Kit (Affymetrix) according to the manufacturer's
 
Hybridization protocol Labeled cDNA was hybridized wiht GeneChip® Hybridization, Wash, and Stain Kit (Affymetrix) as described by the manufacturer.
Scan protocol Affymetrix Human Gene 1.0 ST Array was scanned with the Affymetrix GeneChip Scanner 7G and the FS450_0007 protocol , as recommended in the GeneChip® Hybridization, Wash, and Stain Kit (Affymetrix)
