Treatment protocol Single cell suspension from an luminal-like breast cancer was prepared by enzymatic disaggregation of tumours. The cells were re-suspended in cold staining buffer (PBS containing 0.5 % FCS and 3 % human immune globulin (Gammaguard) (N.V Baxter S.A, Belgium) and stained with fluorescently-labelled antibodies, diluted according to the manufacturers’s recommendation. Following 30 min incubation at 4 oC, the stained cells were washed ones with PBS, then re-suspended in DMEM/F12 with 0.,5% human serum albumin, 2% Hepes, Penicillin and Streptomycin and further sorted by FACS DIVA flow cytometer (Becton Dickinson), equipped with a 488nm Argon laser (Coherent) and 633nm HeNe laser (Spectra Physics).The single cell suspensions were stained with 1 µg/ml propidium iodide (PI) (Sigma) prior to sorting to exclude the dead cells from the analysis.

Extraction protocol Total DNAfrom FACS sorted cell subpopulations was isolated using Qiagen DNA extraction kit according to manufacturers protocol.
