Treatment protocol |
DC were harvested on day 7, incubated with 0.3 µM sulforaphane (Sigma-Aldrich) in DMSO or DMSO alone as control for 2 hours and then activated with 100 ng/ml LPS from E. coli O111:B4 (Sigma-Aldrich) for 1 hour.

Growth protocol |
Bone marrow-derived DC from C57BL/6 mice were isolated and cultivated as described in Lutz MB et al., J Immunol Methods 1999;223:77-92.

DC were lysed and total RNA was isolated using the peqGOLD Total RNA Kit (peqlab) following the manufacturer's protocols.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100ng total RNA using the GeneChip® WT PLUS Reagent Kit (Affymetrix).

Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix MoGene 2.1 ST Array Plates. Hybridization, washing, staining and scanning was performed fully automated on a Gene Titan instrument using GeneChip® Hybridization, Wash, and Stain Kit (Affymetrix).
