Treatment protocol |
Resting or activated umbilical cord blood Tregs were added at day 9 of culture. Resting Tregs were added freshly isolated whereas activated Tregs were pre-activated for 24 h with plate bound CD3, soluble CD28 and IL-2 and washed before being added to HSC cultures.

Growth protocol |
UCB HSC were cultured in the presence of resting or activated Tregs from day 9 until day 12, then HSC were re-isolated using a MoFlow cell sorter.

Extraction protocol |
RNA extraction was performed using the Rneasy mini kit (Qiagen) following manufacturer's recommendations. RNA concentration was measured using the NanoDrop 1000 Spectrophotometer (Thermo Scientific, USA).

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
