Cells proliferated as spheres. After one or two passages, spheres were placed in Trizol reagent.

| Growth protocol |
Epidermis was dissected from dermis following enzymatic digestion. The dermis was cut into small pieces and further digested and filtered until single cells were isolated. These cells were cultured in proliferation-promoting conditions as previously described.

Trizol extraction of total RNA was performed with the RNeasy Mini Kit (Qiagen) according to manufacturer's instructions.

Single-stranded cDNA was generated from the cRNA with the WT cDNA Synthesis Kit (Affymetrix) and then fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).

Samples were hybridized with GeneChip Human Gene 2.0 ST Arrays (Affymetrix) and scanned at the HSC Microarray Facility.

Raw probe intensity values were background corrected, normalized with quantile normalization, transformed into the log2 scale, and summarized into probesets using the Robust Multichip Analysis (RMA) algorithm at the gene level in the Affymetrix Expression Console program.
