Treatment protocol |
A549 cells were transfected with either control siRNA (D-001810-10-05, Dharmacon) or Xrn2 siRNA (L-017622-01-0010, Dharmacon) pools using the Lipofectamine™ RNAiMAX Transfection Reagent (Invitrogen). Transfections were performed according to the manufacturer’s instructions. 48 hours post transfection, the cells were infected with the Influenza A/Puerto Rico/8/34(ΔNS1) strain at an multiplicity of infection of 3. Wells of uninfected cells were used as controls for infection. Cells were collected for further processing 4 hours post infection. RNA and protein levels of Xrn2 were quantified to determine knockdown efficiency. Note: This experiment was done in parallel with the experiments described in GSE52933 and GSE52932 share the same UT and siCtrl treated controls, and hence share the same raw values for those two conditions

Data processing |
Data was analysed using Genespring GX12.5 . To analyze gene expression changes during stimulation, raw data were subject to quantile normalization and baseline transformation was performed to the median of unstimulated samples of the corresponding siRNA treatment.
