Embryonic day 17.5 (E17.5) nasal septum or postnatal day 1 (p1) rib chondrocytes isolated from wild-type ICR mice were subject to Trizol total RNA isolation. Isolated RNAs were digested with DNAse I.

Three hundred ng of total RNA were labeled for each sample using the GeneChip WT cDNA Synthesis and Amplification Kit and WT Terminal Labeling Kit (900673 and 900671; Affymetrix)

Hybridization protocol |
Following fragmentation, 25ul of cDNA were hybridized for 16 hr at 45C on GeneChip Mouse Gene Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Chips were scanned with GeneChip Scanner 3000 (Affymetrix) to generate image and CEL files, using Affymetrix default analysis settings and global scaling as normalization method
