Treatment protocol For estradiol stimulation, cells were grown in phenol red free medium with charcoal stripped serum for several days and treated with 100nM of estradiol for 45 min.
Growth protocol The human breast cancer MCF7 cells (ATCC # HTB-22) were grown in DMEM supplemented with 10% fetal bovine serum (FBS).

Extraction protocol For ChIP-seq, cells were crosslinked using 1% formaldehyde and lysates were clarified from sonicated nuclei and histone-DNA complexes were isolated with antibody.
ChIP-seq: Libraries were barcoded (NEXTflex™ DNA Barcodes) (Bio Scientific, Austin, TX).
