Sample GSM2290155 Query DataSets for GSM2290155
Status Public on Aug 29, 2018
Title HUVEC Notch1 siRNA_2
Sample type RNA

Source name Human umbilical vein endothelial cells
Organism Homo sapiens
Characteristics tissue: Human umbilical vein endothelial cells
sirna: Notch1
stimulation: medium
Treatment protocol Cells were seeded 37.500 cells/cm2, cultured for 24 hours and transfected with siRNA (scrambled or targeting Notch1). 48 hours after transfection, cells were stimulated with recombinant human interleukin-1b (5ng/ml, control cells were given fresh medium) for 4 hours before harvesting in Qiagen RLT lysis buffer.
Growth protocol Pooled HUVEC from 10 donors were cultured in MCDB 131 with 7.5% FBS, 10 ng/ml EGF, 1 ng/ml bFGF, 1μg/ml hydrocortisone, 50 μg/ml gentamicin, 250 ng/ml fungizone and 1% L-glutamine. Cells were maintained at 37°C in human 95% air/ 5% CO2 atmosphere.
Extracted molecule total RNA
Extraction protocol RNA was extracted using the Qiagen RNEasy Mini kit, according to manufacturers instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared with the Illumina® TotalPrep™-96 RNA Amplification Kit

Hybridization protocol Standard Illumina hybridization protocol
Scan protocol Standard Illumina scanning protocol
Description replicate 1
Data processing Raw data (.idat files) were read and processed by R/Bioconductor/beadarray
