Treatment protocol B16-OVA cells were incubated either with BO-112 at 0.5 µg/mL (n=3) or identical volumes of vehicle (5%glucose, n=3). After 24h, cells were collected and total RNA was isolated in two steps using TRIzol (Life technologies) and Rneasy Mini-Kit (Quiagen) purification, following manufacturer`s RNA cleanup protocol.
Growth protocol B12-OVA cells were cultured in RPMI 1640 containing 10% fetal bovine serum, 2mM glutamine, 100 U/ml penicillin and 100 µg/ml streptomycin, and 50 µM 2-mercaptoethanol and 400µg/mL Geneticin (Gibco).
Extracted molecule total RNA
Extraction protocol The cells were harvested with TRIzol Reagent (Invitrogen) and the RNA was extracted according to the manufacturer's instructions. As a last step of the extraction procedure, the RNA was purified with the RNeasy Mini-kit (Qiagen, Hilden, Germany).
Label Biotin
Label protocol The sense cDNA was prepared from 300 ng of total RNA using the Ambion® WT Expression Kit. The sense strand cDNA was then fragmented and biotinylated with the Affymetrix GeneChip® WT Terminal Labeling Kit (PN 900671).
 
Hybridization protocol Labeled sense cDNA was hybridized to the Affymetrix Mouse Clariom S array according to the the manufacturer protocols and using GeneChip® Hybridization, Wash and Stain Kit.
Scan protocol Genechips were scanned with the Affymetrix GeneChip® Scanner 3000.
Description Gene expression data from whole B16-OVA cell line
Data processing Data was normalized using Bioconductor software and RMA algorithm.
