Growth protocol BEAS2B cells were grown in advanced DMEM-F12 media (ThermoFisher, MA) supplemented with 1% fetal bovine serum and 100 U/mL Penicillin, 100 g/mL of Streptomycin, 292 g/mL L-Glutamine and 1% HEPES. All cultures were maintained at 37o C in a humidified 5% CO2 atmosphere and tested to ensure absence of Mycoplasma. Cultures were partitioned before >80% confluencey.
Extracted molecule total RNA
Extraction protocol RNA was isolated using the Rneasy Plus Mini Kit.
Samples were checked using BioA and libraries were prepared using the TruSeq Stranded mRNA kit. Each library was quantified using Qubit and then pooled toether. Library pool was quantified using qPCR. Pool was then diluted to 4 nM and loaded on the HiSeq High Output Mode. Run was performed using a full flow cell (8 lanes) PE 100. 5% PhiX was spiked in to check run qaulity.
