Treatment protocol |
LNCaP cells were plated at 1x10^6 cells per 150 x 25mm dish with 25mL of growth media, and allowed to adhere for 48h prior to treatment. Each treatment was prepared at 6x concentration, so that 5mL of treatment soultion was added to the 25mL growth media.

Growth protocol |
LNCaP cells were grown in RPMI-1640 media with 10% fetal bovine serume supplemented with 100U penicillin and 0.1 mg/mL streptomycin in 37°C with 5%CO2 and 85% humidity.

Total RNA was extracted using the RNeasy Midi kit (Qiagen Inc., Valencia, CA, USA) DNA was removed by on-column DNase digestion. RNA quality and concentration was determined by analysis with an Agilent 2100 bioanalyzer at the Center for Functional Genomics at State University of New York at Albany (Rensselaer, NY, USA)
