Treatment protocol |
All human postmortem brain tissue samples were obtained from the NICHD Brain and Tissue Bank for Developmental Disorders (NICHDBB)(Baltimore, MD, USA). All subjects were defined as normal controls by forensic pathologists at the NICHDBB. No subjects with prolonged agonal state were used.

Extraction protocol |
Total RNA was extracted from 50mg frozen sample using a mirVana RNA Isolation Kit (Ambion, AM1560), according to the manufacturer’s instructions.The quality of each sample was checked by using an Agilent 2100 Bioanalyzer.

For each sample, 100ng total of RNA was dephosphorylated and ligated with pCp-Cy3, following the manufacturer’s instructions, as described in the miRNA Complete Labeling and Hybridization Kit (Agilent Technologies).

The labeled RNA was purified and hybridized to Agilent Human miRNA Microarray following the manufacturer’s instructions

Scan protocol |
The slides were scanned with the Agilent Microarray Scanner using one color scan setting for array slides.

Data processing |
The scanned images were analyzed with Feature Extraction Software (Agilent) to obtain TotalGeneSignal. microRNAs with features in SystematicName column begining with hsa- and in column IsGeneDetected as 1 remained, others were removed. This resulted in detecting expression of 429 human miRNA in at least one of the 9 samples studied.
