Growth protocol The hepatoma cell lines SK-HEP-1 [American Type Culture Collection (ATCC) number HTB-52], HepG2( ATCC number HB-8065), Hep 3B( ATCC number HB-8064), Huh7, MHCC97-H and MHCC97-L, SMMC-7721 (China Center for Type Culture Collection, resource number 3115CNCB00263), and HEK293 and HeLa cell lines were cultured and maintained in Dulbecco’s modified Eagle medium (DMEM) or modified Eagle's minimal essential medium (MEM), supplemented with 100 ml/l fetal calf serum (FCS, Gibco, USA), 100 IU/ml penicillin, 100μg /ml streptomycin at 37 ℃ in a humidified incubator with 5% CO2 (see Supplementary Figure S1). Normal primary human hepatocytes (PHHC-1, PHHC-2, PHHC-3) were isolated from 3 normal liver tissues adjacent to hepatic hemangioma under sterile conditions using a modified four-step retrograde perfusion technique as reported elsewhere.

Extraction protocol The MirVanaTM miRNA Isolation Kit (Ambion, AM1560) was used to isolate total RNA (including miRNA) from all cell lines according to the manufacturer’s protocol.
