Treatment protocol |
Differentiation medium was composed of: DMEM/Ham’s F12 media supplemented with 10 µg/ml transferrin, 0.86 µM insulin, 0.2 nM triiodothyronine, 1 µM dexamethasone, 100 µM isobutyl-methylxanthine and 1 µM rosiglitazone. Three days after first exposure, the medium was changed and dexamethasone and isobutyl-methylxanthine were omitted.

SREK kit by Applied Biosystems (life technologies) Libraries were constructed either with adapter set A (for sequencing microRNA from the 5' to 3' end) or with adapter set B (for sequencing microRNA from the 3' to the 5' end). The sequence of adaptor A and B sequenced in the reads are the same: CGCCTTGGCCGTACAGCAG (330201030313112312 in colorspace). Libraries were size selected on acrylamide gels (110 to 140 nucleotides)
