Treatment protocol |
24 mice were separated evenly into two treatment groups (placebo vs. DSHN). 10 days post-grafting was allowed for metastasis to occur before treatments. Dosage was 10mg/kg/mouse, and mice were treated daily for 7 days. Lungs were collected and 3 lungs from the same treatment group were pooled to be one sample.

Pooled lung samples were processed immediately for dissociation using Lung Dissociation Kit from Miltenyi Biotec (130-095-927). Dendritic cells were isolated from each pooled sample using CD11b (130-126-725) and CD11c (130-125-835) beads from Miltenyi Biotec. RNA was isolated using Qiagen RNeasy Mini Kit

Hybridization protocol |
Hybridization reaction was set up based on the NanoString manuals. We calculated required amounts of RNA samples based on DV200, mix with the NanoString reporter CodeSet, hybridization buffer and Capture ProbeSet. The hybridization master mix was incubated at 65C for 20 hours and then loaded to the cartridge to run the assay.

NanoString nCounter Sprint Profiler was used to examine RNA targets in samples; Mouse Myeloid Innate Immunity V2 was selected.

nSolver Analysis Software 4.0 was the software used for data analysis; data was analyzed according to the user manual
