Treatment protocol co-cultures were treated with : PD 173074 (1mM in DMSO), SB431542 (10μM in DMSO), or 2μl/mL DMSO (control), for 24 hours.
Growth protocol ECs were grown in transwell co-culture with NSPCs,in growth factor-free NSPC medium with addition of 5% FBS, at 37C, in CO2 incubator

Extraction protocol After 24 hours in transwell co-culture, the ECs and NSPCs were separated and miRNA s were isolated using mirVana miRNA (AB/Ambion) isolation kit

Hybridization protocol Following labeling, 21.5 ul of biotin-labeled RNA was hybridized for 16 hr at 48C on Affymetrix miRNA 2.0 Arrays. The miRNA 2.0 Arrays were washed and stained in the Affymetrix Fluidics Station 450 using fludics script FS450_0003.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.
Description miRNA expression data from EC/NSPC co-culture treated with DMSO (control)
Data processing Data files were analyzed with Affymetrix miRNAQCtool using RMA background correction and quantile normalization (Default settings)
