Banxia Xiexin decoction promotes gastric lymphatic pumping by regulating lymphatic smooth muscle cell contraction and energy metabolism in a stress-induced gastric ulceration rat model

(Submitter supplied) The roles of lymphatic pumping disorder in gastrointestinal diseases have drawn increasing research attentions. A Traditional Chinese medicine (TCM) formula Banxia Xiexin decoction (BXD) has definite therapeutic effect in treating stress-induced gastric ulceration (SIGU) and many other gastrointestinal diseases, but its effect on gastric lymphatic pumping (GLP) remains unclear. This study used tracer-aided in vivo imaging to illustrate BXD’s GLP recovery effect in a SIGU rat model, and explored the underlying mechanisms via RNA sequencing (RNA-Seq) and transcriptomic analysis of gastric lymphatic vessel (GLV). more...

Small-molecule-induced epigenetic rejuvenation overcomes myelinogenic barriers and stimulates myelin repair [RNA-seq]

(Submitter supplied) Failure of oligodendrocytes to remyelinate underlies diseases such as multiple sclerosis (MS). We found that epigenetic silencing prevents oligodendrocytes from producing myelin sheaths in demyelinating MS lesions. Here, we developed a transgenic reporter system to identify a small-molecule epigenetic modulator that stimulates oligodendrocyte maturation and myelin ensheathment in vitro. This compound promoted remyelination in animal models of MS and myelination of regenerated axons and increased myelin sheath lengths in human iPSC-derived organoids. more...

Netrin-1 engineered endothelial cell exosomes induce the formation of pre-regenerative niche to accelerate peripheral nerve repair [miRNA-seq]

(Submitter supplied) The formation of vascular niche is pivotal during the early stage of peripheral nerve regeneration. This antecedent angiogenesis meets the nutrient requirements for subsequent rapid axon regeneration and remyelination. Nevertheless, the mechanisms of vascular niche in the regulation of peripheral nerve remain unclear. The axon guidance molecule Netrin-1 (NTN1) is widely expressed in peripheral nerves and particularly was found up-regulated in sciatic nerve stump after peripheral nerve injury (PNI). more...

Defined conditions control the morphological dualism of rat primitive extraembryonic endoderm stem cells

(Submitter supplied) We report two chemically defined conditions that differ by the addition of the cytokine Lif and the β-catenin-stabilizing drug Chir99021 and enable permanent self-renewal as mesenchymal and epithelial morphotypes, respectively. The morphotypes are interconvertible and differentiate equipotently. Surprisingly, the proliferation of both morphotypes requires Lif/Gp130/Stat3 signaling (autocrine in the absence of added Lif) and non-canonical Wnt signaling (autocrine). more...

Study of the tRNA-derived fragment expression profile in rat pancreatic islets of diabetes-susceptible low-protein (LP) exposed rats

(Submitter supplied) In this study, we discovered cytosolic and mitochondrial fragments resulting from tRNA and mt-tRNA cleavage, which may act as new regulators of cellular and metabolic functions. The so-called LP rat model consists of 22-day-old rats whose mothers have been subjected to a low protein diet during gestation and lactation. These offspring have impaired pancreatic development with reduced ß-cell mass, glucose-stimulated insulin secretion defect and are more prone to develop diabetes in adulthood. more...

A systems-level analysis of bile acids effects on rat colon epithelial cells

(Submitter supplied) Bile acid diarrhoea is a chronic condition caused by increased delivery of bile acids to the colon. The underlying mechanisms remain to be elucidated. To investigate genes involved in bile acid diarrhoea, systems-level analyses were employed on a rat bile acid diarrhoea model. Twelve male Wistar Munich rats, housed in metabolic cages, were fed either control or bile acid-mixed (1% w/w) diets for ten days. more...

Next generation sequencing for miRNA profile of blood sample of goiter rat model treated by quemeiteng granule

(Submitter supplied) MicroRNAs play important roles in the regulatory processes of occurrence and progression of goiter. In the present study, whole transcriptome sequencing analysis was applied to investigate the expression profiles of miRNAs. After comparing blood samples of propylthiouracil (PTU)-induced goiter model blood samples and quemeiteng granule (QMTG) treated groups, 1 miRNA was found to be differentially expressed. more...

Comparison of transcriptomic profiles between HFPO-DA and positive controls for PPARa, PPARg or cytotoxicity in mouse, rat, and pooled human hepatocytes

(Submitter supplied) Like many per- or polyfluorinated alkyl substances (PFAS), toxicity studies with HFPO-DA (ammonium, 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy)-propanoate; CASRN 62037-80-3), a short-chain PFAS used in the manufacture of some types of fluorinated polymers, indicate that the liver is the primary target of toxicity in rodents following oral exposure. Although the current weight of evidence supports the PPARa mode of action (MOA) for liver effects in HFPO-DA-exposed mice, alternate MOAs have also been hypothesized including PPARg or cytotoxicity. more...

Effects of silencing miR-142-3p on the pathological and physiological functions of cardiac fibroblasts

(Submitter supplied) MicroRNAs (miRs), a group of small and non-coding RNAs, negatively regulate gene expression via promoting messenger RNA (mRNA) degradation or blocking mRNA translation. Many miRs have been recognized as biomarkers or possible targets for the diagnosis or therapy of some diseases. Among them, miR-142-3p was involved in the occurrences and progression of various cardiovascular diseases. Previous studies found that miR-142-3p upregulation could ameliorate myocardial ischemia/reperfusion (I/R)-induced transdifferentiation of fibroblasts to myofibroblasts and collagen deposition. more...

Role of MYC acetylated lysine residues in cell transformation and gene-selective regulation [Rat1A]

(Submitter supplied) The functions of MYC acetylation have remained unclear. In this study we identified the major lysine residues of MYC that are acetylated by p300 and GCN5 in cancer cell lines and established the requirment of three specific acetyl-lysine (AcK) residues for MYC transformation of Rat1a fibroblasts and MCF10 mammary epithelial cells. We further uncovered gene-selective regulatory functions and conserved genetic programs and biolofical processes regulated by MYC AcK residues in both cell types.

Species-specific responses during Seoul orthohantavirus infection in human and rat lung microvascular endothelial cells

(Submitter supplied) Orthohantaviruses are viruses generally carried by rodents in which they do not cause overt disease. Seoul orthohantavirus (SEOV) predominantly exists as a persistent infection in the omnipresent reservoir host, the Norway rat, Rattus norvegicus. Upon respiratory transmission via aerosolized infectious rodent excreta to humans, SEOV causes an acute disease named hemorrhagic fever with renal syndrome (HFRS). more...

ATAC and histone H3K9me3 landscapes reveal long-term epigenomic effects of fetal-neonatal iron deficiency in rat hippocampus [ChIP-seq]

(Submitter supplied) Background: Iron deficiency (ID) during the fetal-neonatal period results in long-term neurodevelopmental impairments associated with pervasive hippocampal gene dysregulation. Prenatal choline supplementation partially normalizes these effects, suggesting an interaction between iron and choline in hippocampal transcriptome regulation. To understand the regulatory mechanisms, we investigated epigenetic marks of genes that are poised to be activated (ATAC-seq) or repressed (H3K9me3 ChIP-seq) in iron-repleted adult rats having experienced fetal-neonatal ID exposure with or without prenatal choline supplementation. more...

Rat models for Portosinusoidal Vascular Disease

(Submitter supplied) Background: Porto-Sinusoidal Vascular Disorder (PSVD) involves a group of rare vascular liver diseases of unknown etiology that leads to the development of portal hypertension and its life-threatening complications. Its pathophysiology is not well understood and animal models described to date do not fully recapitulate human disease. Methods: We developed 3 different PSVD rat models by either immunosensitization (repetitive intraportal LPS or intramuscular spleen extract injections) or toxic (combination of FOLFOX and a selenium-enriched diet, Selfox model) treatment and characterized them at hemodynamic, histological, biochemical and transcriptional levels. more...
