ChIP-Seq analysis to identify direct binding of ZNF165

(Submitter supplied) We found that the cancer testis antigen, ZNF165, is required for viability and modulates TGFβ-induced gene expression in mesenchymal, Claudin-Low, TNBC. To begin to define ZNF165's role in TNBC tumorigenesis, we performed ChIP-Seq analysis in the mesenchymal TNBC tumor derived cell line, WHIM12, stably expressing ZNF165-V5. This analysis uncovered 381 peaks associated with 410 genes and included TGFβ target genes, SMURF2 and SMAD, that promote negative TGFβ feedback regulation. more...

Expression data to identify putative transcriptional targets of ZNF165 in Triple Negative Breast Cancer (TNBC)

(Submitter supplied) We found that the cancer testis antigen, ZNF165, is required for viability and can modulate TGFβ-induced gene expression in mesenchymal, Claudin-Low, TNBC. We employed the Affymetrix microarray platform to uncover transcriptionally modulated genes following ZNF165 depletion and TGFβ stimulation using the Claudin-low TNBC tumor-derived cell lines, SUM159 as a model. Our results provide insight into how ZNF165 globally modulates TGFβ signaling. more...

Genomic binding profiles for ZNF165, ZNF446, and SMAD3 in triple-negative breast cancer

(Submitter supplied) The testis protein ZNF165 is aberrantly expressed in triple-negative breast cancer (TNBC), where it modulates TGFβ signaling to promote tumor growth and survival. To investigate whether ZNF165 functions in this capacity via cooperation with SMAD3, a canonical TGFβ-induced transcription factor, we performed ChIP-seq for both factors in TNBC cell lines (WHIM12 and SUM159). Additionally, we established genomic binding profiles for ZNF446, a previously uncharacterized transcription factor that we have identified as a member of a ZNF165-SMAD3 complex. more...

Down-regulation of the TGF-b pseudoreceptor BAMBI enhances TGF-b signaling and promotes invasion of Non-Small Lung Cancer: DNA methylation analysis

(Submitter supplied) Genome wide DNA methylation profiling of lung cancer tissue samples and corresponding normal lung tissue samples of 40 patients. The Illumina Infinium 450k Human DNA methylation Beadchip was used to obtain DNA methylation profiles across approximately 480,000 CpGs. Samples included 40 normal lung tissue samples and 40 lung cancer samples.

Gene expression profiles in CDX2P-G19Cre;Apcflox/flox;Tgfbr2flox/flox and CDX2P-G19Cre;Apcflox/flox mouse tumors

(Submitter supplied) Mutations in TGFBR2, a component of the transforming growth factor (TGF)-β signaling pathway, occur in high-frequency microsatellite instability (MSI-H) colorectal cancer (CRC). In mouse models, Tgfbr2 inactivation in the intestinal epithelium accelerates the development of malignant intestinal tumors in combination with disruption of the Wnt-β-catenin pathway. However, no studies have further identified the genes influenced by TGFBR2 inactivation following disruption of the Wnt-β-catenin pathway. more...

REV1 promotes lung tumorigenesis by activating the Rad18/SERTAD2 axis

(Submitter supplied) REV1 is the central member of the family of TLS polymerases, which participate in various DNA damage repair and tolerance pathways, including translesion DNA synthesis, and play a significant role in maintaining genomic stability. However, the role of REV1 in tumors is rarely reported. In this study, we found that the expression of REV1 was significantly upregulated in lung cancer tissues compared with matched adjacent tissues and was associated with poor prognosis. more...
