Title |
JAK2 mutant |
Sample type |
RNA |
Source name |
JAK2 mutant
Organism |
Homo sapiens |
Characteristics |
cell line: KBM7 chronic myeloid leukemia (CML) cells genotype/variation: JAK2 mutant agent: unstimulated
Treatment protocol |
Log phase proliferating cells were stimulated with IFN-gamma (100ng/ml, Peprotech) or TNF-alpha (10ng/ml, Peprotech) for 6 hours or left untreated. Sample WT_1 corresponds with the control sample for the IFN-gamma stimulation; Sample WT_2 corresponds with the control sample for the TNF-alpha stimulation.
Growth protocol |
KBM7 cells were maintained in Iscove’s Modified Dulbecco’s Medium (IMDM) supplemented with 10% fetal calf serum and 1% penicillin/streptomycin.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was isolated with the RNeasy Mini Kit from Qiagen with the help of the QIAcube robot.
Label |
Biotin
Label protocol |
100ng of total RNA were amplified and labeled using the Affymetix Genechip 3` IVT Express Kit (Catalog Number 901228, Affymetrix, Santa Clara, CA)
Hybridization protocol |
Affymetrix GeneChip® Human PrimeView arrays were washed, stained and scanned according to the protocol described in GeneChip® 3` IVT Express Kit User Manual (Fluidics Protocol FS450_0002)
Scan protocol |
Scanning was done on an Affymetrix GeneChip Scanner 3000 7G, software AGCC 3.1.1
